|Abstract:||Biochip technology could allow rapid multiparametric diagnosis of infectious diseases and related antimicrobial resistance genes. Two aspects of biochips were studied during this project. First, to facilitate DNA or PNA capture probes design, we studied the impact on hybridisation signal of the position targeted by the probe. We observed that the length of the target’s non-hybridised overhangs had an influence on obtained hybridisation signal and that it could be responsible for false-negatives. Second, in order to contribute to the elimination of sample nucleic acids amplification and labelling before hybridisation, we developed a biochip made of PNA probes immobilised on a glass slides. This biochip allows detection using a soluble fluorescent cationic polythiophene that acts as hybridisation transducer.|
